Using 240bp fragments with either A overhangs or T overhangs as substrates, reactions were carried out at 16℃ for 10, 20, 30, and 40 minutes in 5×T4 DNA Ligase Buffer (for NGS). Compared to Vendor A's and Vendor B's T4 DNA Ligase, the equimolar amount of QuarPro Superfast T4 DNA Ligase from Dynegene showed a higher ligation efficiency at the same time points.

Using 240 bp fragments with either A overhangs or T overhangs as substrates, reactions were carried out at 16℃ for 30 minutes in 5×T4 DNA Ligase Buffer (for NGS). The self-produced QuarPro Superfast T4 DNA Ligase from Dynegene showed a TA ligation efficiency of 86.4±5.0%, while the equimolar amounts of Vendor A's and Vendor B's T4 DNA Ligase showed TA ligation efficiencies of 57.4±5.0% and 34.2±5.0%, respectively.
